The stratum corneum is the outermost layer of the epidermis, functioning as the body's primary interface with the external environment and its main permeability barrier (StatPearls: Physiology, Stratum Corneum, 2023). It is structurally characterized by the bricks and mortar model, where protein-rich corneocytes (the bricks) are embedded in a continuous matrix of intercellular lipids (the mortar), primarily ceramides, cholesterol, and free fatty acids (Journal of Clinical Medicine, 2018). Drugs like petrolatum act as occlusives to prevent water loss, while humectants like glycerin draw water into the stratum corneum, and keratolytics like salicylic acid promote the shedding of dead cells by disrupting intercellular adhesions (StatPearls: Keratolytic Agents, 2023). Because this target represents a complex tissue layer rather than a single molecular entity, it is often considered a non-specific pharmacological target where the goal is the restoration of physical and chemical homeostasis.
