Overview

Non-structural maintenance of chromosomes element 3 homolog, abbreviated as NSMCE3, is a protein component of the SMC5-SMC6 chromatin organizing complex and belongs to the MAGE superfamily[3][4][7][8]. The SMC5-SMC6 complex is essential for DNA double-strand break repair by homologous recombination, contributing to genome stability, chromosome segregation, and telomere maintenance, especially via the ALT (alternative lengthening of telomeres) pathway[1][3][8]. NSMCE3 modulates sumoylation activities (particularly of shelterin complex proteins), influences ubiquitin ligase activity, and may function as a growth suppressor by facilitating cell cycle arrest[1][3][8]. Pathogenic mutations in NSMCE3 are known to cause syndromes characterized by immunodeficiency and chromosome instability, and dysregulation may contribute to cancer development, especially those associated with telomere maintenance defects[3][6][7]. No drugs directly target NSMCE3, and it is not currently considered a therapeutic target, but its function is central to pathways critical for DNA repair and genome maintenance.
