Overview

The Non-typeable Haemophilus influenzae (NTHi) protein E–pilin A fusion protein (PE-PilA) is a chimeric recombinant antigen designed for use in vaccines targeting respiratory infections (Wilkinson et al., 2017, PubMed). Protein E is a highly conserved 16 kDa surface lipoprotein that functions as an adhesin, binding to vitronectin to facilitate host cell attachment and providing serum resistance by inhibiting the complement system (Hallström et al., 2009, Journal of Immunology). Pilin A is the major subunit of Type IV pili, which are critical for NTHi adherence, twitching motility, and the formation of stable biofilms (Bakaletz et al., 2005, Infection and Immunity). By fusing these two conserved proteins, the vaccine candidate aims to elicit a broad and synergistic immune response (Pichichero, 2015, Expert Review of Vaccines). This response includes the production of antibodies that block bacterial colonization and enhance opsonophagocytic killing, thereby reducing the incidence of NTHi-related conditions such as acute otitis media in children and exacerbations of chronic obstructive pulmonary disease (COPD) in adults (Wilkinson et al., 2019, Lancet Infectious Diseases).
