Non-VCAN mRNAs refers to the collective set of messenger RNA transcripts within a cell that do not encode the Versican (VCAN) protein. This term is not a specific therapeutic target but rather a category used in the context of RNA-targeted drug discovery, such as antisense oligonucleotides (ASOs) or small interfering RNAs (siRNAs), to define the off-target landscape. Versican is a critical extracellular matrix proteoglycan involved in cell adhesion, proliferation, and migration, and its dysregulation is linked to cancer, cardiovascular disease, and inflammation (UniProt P13611). When designing therapies to modulate VCAN, it is essential to ensure high specificity to avoid binding to non-VCAN mRNAs, which could lead to unintended gene silencing and cellular toxicity (Nature Reviews Drug Discovery, 2020). Consequently, non-VCAN mRNAs represent the negative selection criteria used during bioinformatics screening and lead optimization to ensure drug safety and minimize side effects.
