Overview

Nonspecific protein binding (NSB) refers to the unintended association of a molecule, such as a drug or chemical probe, with proteins or surfaces other than its primary biological target (IUPHAR/BPS Guide to Pharmacology). This phenomenon is typically mediated by non-covalent interactions, including hydrophobic effects, electrostatic forces, and van der Waals interactions, and is generally characterized as a low-affinity, high-capacity process (Journal of Pharmaceutical Sciences, 2010). In a clinical context, nonspecific binding to plasma proteins like albumin or alpha-1-acid glycoprotein can sequester drugs, thereby reducing the free fraction available to exert therapeutic effects (StatPearls, 2023). In laboratory settings, it often presents a significant challenge by increasing background noise in assays, which can lead to inaccurate measurements of binding affinity or potency (Nature Methods, 2013). Because it is a physical property of a molecule's interaction with its environment rather than a specific biological entity, it is not considered a therapeutic target. Consequently, managing nonspecific binding is a critical aspect of drug design, pharmacokinetic modeling, and the development of sensitive diagnostic tools.
