Overview

Nonstructural protein 1 (NS1) of Zika virus is a glycoprotein of approximately 46–55 kDa, multifunctional in the viral life cycle and a key pathogenic factor[1][3]. It is present in different oligomeric forms: as an intracellular dimer essential for viral RNA replication, as a membrane-associated form interacting with viral replication complexes, and as a secreted hexamer or tetramer involved in immune evasion and pathogenesis[1][2][4][5]. NS1 mediates immune evasion by interfering with host immune responses and serves as a major antigenic marker for infection. NS1’s surfaces exhibit unique electrostatic properties among flaviviruses, potentially underlying diverse pathogenic mechanisms and interactions with host proteins. Variants of NS1, its glycosylation status, and its induced antibody responses are implicated in both protective immunity and pathology, including possible autoimmunity during infection or after immunization[1][3][4][5][6]. NS1 is under investigation as a diagnostic biomarker, immune target, and potential vaccine antigen against Zika and related viruses.
