Overview
Papain-like protease (PLpro) is a multifunctional cysteine protease found in coronaviruses, including SARS-CoV, MERS-CoV, and SARS-CoV-2. It is an essential viral enzyme responsible for processing the viral polyprotein and modulating host immune responses. PLpro is encoded as part of nonstructural protein 3 (nsp3), which is produced from the large replicase polyprotein 1a/1ab. PLpro functions as a viral protease and also possesses deubiquitinating and deISGylating activities, allowing it to interfere with host immune signaling.
