Nontypeable Haemophilus influenzae type IV pilin subunit A (PilA) is a hydrophobic protein that polymerizes to form the core structure of type IV pili on the surface of NTHi. These pili are critical for the bacterium's ability to adhere to host tissues, form biofilms, and establish chronic infections. In vivo, PilA is involved in the stability and structural integrity of the biofilm matrix, making it a key virulence factor. Disruption of PilA, such as through genetic knockout, results in significantly impaired bacterial adhesion and less robust biofilm formation[2][3].
