Overview

NOP58 ribonucleoprotein (NOP58) is a central, highly conserved core component of box C/D small nucleolar ribonucleoprotein (snoRNP) complexes, which are essential for the processing and modification of ribosomal RNA (rRNA)[2][3][1]. NOP58 is fundamental to the biogenesis and function of box C/D snoRNAs (such as U3, U8, U14), and plays an indispensable role in the nucleolar processing of pre-rRNA, especially mediating 2'-O-methylation—a key modification that influences ribosome function and, by extension, protein synthesis fidelity[3][4]. NOP58 operates as a scaffold and RNA-binding protein within snoRNPs, interacting primarily with other core proteins like NOP56 and fibrillarin, and is required for correct assembly and optimal enzymatic activity of the complex[1]. Posttranslational modification by SUMOylation is necessary for its high-affinity snoRNA binding and proper snoRNP complex assembly[2]. Dysfunction or dysregulation of NOP58 is linked to various developmental disorders, and altered expression has prognostic implications in certain cancers, possibly by modulating ribosome biogenesis via snoRNA-dependent RNA modifications[3][4]. There are currently no known drugs directly targeting NOP58, and it is not considered a classic therapeutic target or receptor, but rather a core ribonucleoprotein essential for RNA metabolism and ribosome assembly.
