Norepinephrine (NE) release is a crucial physiological process involving the exocytosis of NE-containing vesicles into the synaptic cleft. It is tightly regulated by presynaptic receptors (alpha2 and beta adrenoceptors) and influenced by neuronal activity. NE release is essential for mediating stress responses, arousal, attention, and cardiovascular function. Dysregulation of NE release is implicated in various disorders, including depression, anxiety, and hypertension. Drugs targeting adrenergic receptors or NE reuptake/metabolism can significantly impact NE release and its downstream effects.
Norepinephrine release is modulated by various drugs. Alpha2-adrenergic agonists inhibit NE release through negative feedback. Alpha2-adrenergic antagonists increase NE release by blocking this feedback. Beta-adrenergic agonists can facilitate NE release. Reuptake inhibitors prevent the reuptake of NE, increasing its concentration in the synaptic cleft. MAOIs inhibit the degradation of NE, increasing its availability for release. Amphetamines promote NE release.
