Overview

Normal host tissues represent the healthy, non-diseased cellular components and organ systems of a patient that are not the intended focus of a therapeutic intervention. In pharmacology, these tissues are critical because they are the primary sites where adverse drug reactions (ADRs) and dose-limiting toxicities occur, often defining the safety profile and therapeutic index of a drug (National Cancer Institute, 2024). For instance, while chemotherapy targets rapidly dividing cancer cells, it frequently damages normal host tissues with high turnover rates, such as the bone marrow and intestinal lining (PubMed, 2023). In the realm of advanced biologics like CAR-T cell therapy, normal host tissues may express the target antigen at low levels, leading to 'on-target, off-tumor' toxicity that can result in severe organ damage (Morgan et al., 2010). Understanding the distribution and vulnerability of normal host tissues is essential for the design of targeted delivery systems and the mitigation of systemic side effects in clinical practice (NIH, 2023).
