The Norovirus 3C-like protease (3CLpro), also known as NS6 protease, is a cysteine protease essential for norovirus replication. It cleaves the viral polyprotein into functional non-structural proteins, making it a crucial target for antiviral drug development. Inhibition of this protease blocks viral replication. The enzyme has a chymotrypsin-like fold with two domains and a catalytic dyad composed of Cys139 and His30. Extensive efforts are focused on developing small-molecule inhibitors that bind to the active site and block enzymatic activity.
