Overview

Norovirus capsid protein VP1 (GI.1 genotype) is the main structural component forming the viral capsid of human noroviruses, particularly the prototype Norwalk virus of genogroup I, genotype 1. VP1 assembles into an icosahedral shell with T = 3 symmetry and comprises two key domains: the highly conserved shell (S) domain and the variable protruding (P) domain, which is further divided into P1 and the hypervariable P2 subdomains. The P2 subdomain at the capsid surface mediates binding to host cell histo-blood group antigens (HBGAs) and contains major neutralizing epitopes[1][2][3][4][5]. VP1’s structural flexibility influences antigenic variation and immune escape, posing challenges for vaccine design. Recombinant VP1 assembles into virus-like particles (VLPs) used in vaccine development, but rapid viral evolution and strain diversity can impact vaccine effectiveness. The protein's structural and antigenic features make it a primary focus for therapeutics and diagnostics in norovirus infection[1][3][4][5].
