Overview

Norovirus Genogroup I Genotype 1 (GI.1) Major Capsid Protein VP1 is the primary structural protein of the Norwalk virus, the prototype strain of the Norovirus genus (UniProt P03555). VP1 is composed of approximately 530 amino acids and self-assembles into icosahedral virus-like particles (VLPs) that are morphologically and antigenically similar to the native virus (Prasad et al., 1999). The protein is divided into two main domains: the Shell (S) domain, which forms the inner core, and the Protruding (P) domain, which extends outward and mediates host cell attachment by binding to Histo-Blood Group Antigens (HBGAs) (Tan & Jiang, 2005). Because it is the most immunogenic component of the virus, GI.1 VP1 is the central target for vaccine development, including VLP-based candidates like HIL-214 (Atmar et al., 2011). Vaccines targeting this protein aim to induce neutralizing antibodies that block the interaction between the P domain and host receptors, thereby preventing viral entry and subsequent gastroenteritis. Therapeutic challenges include the high genetic diversity of noroviruses and the potential for antigenic drift, which necessitates the inclusion of multiple genotypes in vaccine formulations to ensure broad protection (Lindesmith et al., 2012).
