Overview

Norovirus GII.4 major capsid protein VP1 is the principal structural protein encoded by ORF2 in human noroviruses of genogroup II, genotype 4. VP1 features a modular architecture with an N-terminal arm (NTA), shell (S) domain forming the icosahedral shell, and a protruding (P) domain divided into P1 and P2 subdomains, distinguished by a flexible hinge region. The P2 subdomain is the most exposed and variable, mediating host attachment by binding to histoblood group antigens and containing antigenic determinants for neutralizing antibodies and immune escape. VP1 assembles into T=3 or T=4 icosahedral capsids in virus-like particles and native virions. GII.4 VP1's conformational and sequence variability drive frequent emergence of antibody escape mutants and epidemic variants, making it a critical target for norovirus vaccine development and molecular epidemiology.
