Overview

The Norovirus virion is the infectious particle of the Norovirus genus, a member of the Caliciviridae family and the leading cause of epidemic non-bacterial gastroenteritis worldwide (CDC, 2023). It is a small, non-enveloped virus characterized by an icosahedral capsid surrounding a single-stranded, positive-sense RNA genome of approximately 7.5 kb (NCBI, 2022). The capsid is primarily composed of the major capsid protein VP1, which facilitates attachment to host cell receptors, specifically histo-blood group antigens (HBGAs), and is the primary target for neutralizing antibodies (UniProt, 2024). Therapeutic strategies focusing on the virion include the development of virus-like particle (VLP) vaccines and small-molecule inhibitors targeting essential viral enzymes such as the RNA-dependent RNA polymerase (RdRp) and the 3C-like protease (PubMed, PMID: 31438526). Despite its significant global health burden, there are currently no FDA-approved vaccines or specific antiviral therapies, largely due to the virus's high genetic diversity and the challenges associated with long-term immune protection (StatPearls, 2023). The virion's extreme environmental stability and low infectious dose further complicate control measures in clinical and community settings.

Mechanism of action

Neutralization of viral capsid proteins (VP1) to prevent host cell attachment, inhibition of viral RNA-dependent RNA polymerase (RdRp) to block genome replication, and inhibition of viral 3C-like protease (NS6) to prevent polyprotein processing (PubMed, PMID: 30634451).
