Overview

The Notch 4 receptor is a type I transmembrane protein and a member of the Notch family of receptors. It plays a critical role in cell fate decisions during development and in adult tissues. NOTCH4 acts as a receptor for membrane-bound ligands, triggering proteolytic cleavage and release of its intracellular domain to modulate gene expression in the nucleus. Dysregulation of NOTCH4 has been associated with susceptibility to schizophrenia, hematologic cancers, and solid tumors, including breast and pancreatic cancer.

Mechanism of action

Ligand binding to NOTCH4 triggers proteolytic cleavage and release of its intracellular domain, which translocates to the nucleus to modulate gene expression via the CBF1/RBPJκ complex.
