Overview

Notch homolog 2 N-terminal-like protein C (NOTCH2NLC) is a human-specific paralog of the canonical Notch receptors, belonging to a gene family that includes NOTCH2NLA and NOTCH2NLB[1][2][5]. It functions primarily as a modulator of the Notch signaling pathway, enhancing Notch receptor activation (NOTCH2, NOTCH1, and NOTCH3) and delaying differentiation of neuronal progenitor cells[2][5]. NOTCH2NLC is expressed in the developing human neocortex, especially in stem cell populations like outer radial glia, and plays a crucial role in increasing the number of cortical stem cells and, consequently, overall cortical neuron number and brain size[1][2][5]. While not a drug target or classical receptor, alterations such as copy number variations or repeat expansions in NOTCH2NLC are associated with neurodevelopmental and neurodegenerative diseases, including neuronal intranuclear inclusion disease (NIID)[5]. NOTCH2NLC is not known to be targeted by any drugs, and it is not classified under classical therapeutic target families like receptors, enzymes, or transporters.
