Overview

NOTCH2NLR, more accurately related to the human-specific NOTCH2NL gene family (including NOTCH2NLA, NOTCH2NLB, and NOTCH2NLC), encodes proteins structurally similar to the N-terminal, EGF-like repeat-rich domain of the NOTCH2 receptor. These proteins are expressed in neural progenitor regions of the developing human cortex and can physically interact with NOTCH2 and other NOTCH receptors, enhancing Notch pathway signaling and promoting maintenance and proliferation of neuronal progenitors, thus delaying their differentiation[2][3]. The "R" designation or "Notch 2 N-terminal like R" is uncommon in major gene/protein databases and likely represents a recently identified paralog, truncated variant, or potentially a pseudogene entry. While NOTCH2NL genes broadly have evolutionary and developmental significance, NOTCH2NLR itself is not recognized as a therapeutic target, and no known drugs or clinical biomarkers are associated with it[2][3].
