Overview

The Notch receptor is a single-pass transmembrane protein that plays a central role in the Notch signaling pathway, regulating cell fate decisions during development and maintaining tissue homeostasis. In mammals, there are four distinct Notch receptors: NOTCH1, NOTCH2, NOTCH3, and NOTCH4. Activation occurs via juxtacrine signaling with ligands like Delta-like and Jagged proteins, leading to proteolytic cleavages and release of NICD, which regulates gene expression. Dysregulation or mutation of Notch receptors is implicated in various diseases, including cancer and developmental disorders.
