Overview

Neurogenic locus notch homolog protein 4 (NOTCH4) is a member of the NOTCH family of receptors. It is a type I transmembrane protein characterized by an extracellular domain with multiple epidermal growth factor-like repeats and an intracellular domain with several distinct motifs. As a receptor for membrane-bound ligands such as Jagged1, Jagged2, and Delta1, NOTCH4 plays a crucial role in regulating cell-fate decisions through the evolutionarily conserved Notch signaling pathway. Upon ligand binding and activation, the released intracellular domain forms a transcriptional activator complex that influences gene expression related to differentiation, proliferation, and apoptosis programs. This receptor is implicated in developmental processes—especially vascular morphogenesis—and has been associated with diseases including hematologic cancers and schizoaffective disorder[1][2]. There is also evidence for its involvement in immune modulation during severe lung inflammation caused by viral infections[9].
