The Notch transcriptional activation complex (NTC) is a multi-protein nuclear complex that serves as the terminal effector of canonical Notch signaling. Upon activation by ligand-induced proteolytic cleavage, the Notch intracellular domain (NICD) is released from the membrane and translocates to the nucleus, where it forms a tripartite complex with the DNA-binding protein CSL (also known as RBPJ in mammals) and the coactivator Mastermind-like (MAML1–3). The NTC binds regulatory DNA elements of Notch target genes and recruits additional chromatin-modifying coactivators (such as p300/CBP, PCAF, and SWI/SNF complexes), resulting in transcriptional activation of genes that control cell fate decisions, proliferation, differentiation, and apoptosis. Dysregulation of the NTC is strongly linked to a range of human disorders, most notably developmental syndromes and cancer, making it a validated therapeutic target but also posing significant toxicity risks for systemic inhibition[1][2][3][4][5][6][7][8].
Inhibitors block formation of the Notch intracellular domain (NICD), thus preventing the assembly of the NTC and transcription of Notch-regulated genes[1][2][3]. Some drugs disrupt NICD–CSL–MAML complex formation directly or indirectly[1][3][5].
