Neurogenic locus notch homolog protein 1 (NOTCH1) is a single-pass transmembrane receptor that plays critical roles in cell fate determination during development and tissue homeostasis. Upon ligand binding, proteolytic cleavage releases the Notch intracellular domain (NICD), which translocates to the nucleus and regulates target gene expression. Aberrant NOTCH1 signaling is implicated in cancer and other developmental disorders.
