Overview

NALT1 (NOTCH1 associated long non-coding RNA in T cell acute lymphoblastic leukemia 1) is a long non-coding RNA gene located upstream of NOTCH1 within 10 kb, identified as playing an oncogenic role in pediatric T-ALL[3][1]. It is upregulated in T-ALL patient samples, and its increased expression correlates with upregulation of NOTCH1. NALT1 is nuclear-localized and acts as a transcriptional activator for the NOTCH1 gene, thereby enhancing NOTCH1 pathway activity and promoting leukemic cell proliferation[1]. Downregulation of NALT1 decreases both NOTCH1 expression and proliferation of T-ALL cells. Targeting the NOTCH signaling pathway through gamma secretase inhibitors reduces the proliferation promoted by NALT1, indicating that NALT1 promotes leukemogenesis predominantly through activation of the NOTCH1 signaling cascade[1]. NALT1 is therefore considered a key regulatory lncRNA in the NOTCH1 pathway and an emerging molecular target for pediatric T-ALL therapy.
