Overview

The Notch1 receptor is a single-pass transmembrane protein and canonical member of the Notch receptor family, functioning as a cell-surface receptor that mediates direct cell-cell communication essential for the regulation of cell fate decisions during embryonic and adult development. Notch1 is activated when neighboring cells present membrane-bound ligands (such as Jagged1, Jagged2, and DLL1), leading to a proteolytic cascade that releases the Notch1 intracellular domain (NICD). NICD translocates to the nucleus where it acts as a transcriptional coactivator with the CSL family of transcription factors and Mastermind-like (MAML) proteins to control target gene expression. Notch1 signaling controls key processes such as differentiation, proliferation, apoptosis, and maintenance of stem and progenitor cells, and dysregulation of this pathway is implicated in various cancers, cardiovascular diseases, and developmental syndromes.
