Overview

The Notch3 receptor is a single-pass transmembrane receptor of the Notch signaling family that plays a crucial role in regulating the development and fate of various cell lineages, particularly in the nervous and vascular systems. In neural crest cells, activation of Notch3 signaling directs the differentiation into mural cell and smooth muscle lineages during embryogenesis and tissue repair. Canonical NOTCH signaling involves ligand-mediated cleavage, nuclear translocation of the NOTCH3 intracellular domain, and transcriptional activation of target genes. Dysregulation of Notch3 signaling has been implicated in several disease processes, including CADASIL (an inherited vascular dementia), vascular malformations, and multiple cancer types where it can regulate cancer stemness and proliferation[1][3][4][5][6].
