Overview

Notchless protein homolog 1 (NLE1) is a WD-repeat-containing protein that acts as a regulatory factor in the Notch signaling pathway and is implicated in controlling the expression of cell cycle and proliferation genes, such as CDKN1A and members of the Wnt pathway. It is normally localized to the nucleolus and nucleoplasm and is required for embryonic development and stem cell survival. NLE1 is overexpressed in several human cancers, where it acts as a tumor promoter by accelerating cell proliferation and suppressing apoptosis, frequently through activation of E2F1-mediated transcription of cell cycle regulators like CDK1 and activation of the PI3K/Akt pathway. NLE1 also serves as a diagnostic and prognostic biomarker for hepatocellular carcinoma and is under investigation as a novel therapeutic target, with some evidence supporting sensitivity to specific small molecule inhibitors.
