Notochord homeobox (NOTO) is a conserved homeobox transcription factor gene that encodes a protein crucial for the proper formation and patterning of the notochord during vertebrate embryonic development[1][2][4][5]. NOTO acts as a master regulator of ciliogenesis in the posterior notochord, modulating the expression of a network of downstream genes (e.g., FOXJ1, RFX3) required for cilia assembly, motility, and the establishment of embryonic left–right axis. Mutation or loss of NOTO leads to severe developmental defects, particularly affecting node morphogenesis, cilia structure, and left–right asymmetry of internal organs, and is essential for specifying axial mesoderm cell fate in the embryo[1][3][4][5]. NOTO’s role is established in mammalian, amphibian, and fish embryos; orthologous genes are present in all vertebrates.
