Novel transcript, antisense to titin" (ENSG00000270277) is a long non-coding RNA identified in the human genome, transcribed from the antisense strand of the Titin (TTN) gene locus on chromosome 2[1][2][3][6]. Antisense lncRNAs at the titin locus are increasingly recognized for their regulatory roles in TTN mRNA splicing and potentially in cardiac physiology[5]. Disruption or modulation of certain antisense transcripts may affect titin protein isoform expression and sarcomere function, especially in heart failure and cardiomyopathy[4][5]. However, there is presently no clinical evidence supporting this transcript as a validated therapeutic target, biomarker, or as being associated with specific disease roles, though ongoing research in RNA regulation in cardiomyopathy suggests emerging potential in RNA-based intervention for titin-related pathologies[4][5].
