NSL1 is a structural protein component of the MIS12 complex, a conserved core assembly at the kinetochore, which is essential for connecting chromosomes to spindle microtubules during mitosis. NSL1, together with other MIS12 components (DSN1, MIS12, NNF1), facilitates the structural bridge between inner centromeric chromatin and the outer kinetochore, mainly through interactions with the NDC80 and KNL1 complexes. NSL1 features coiled-coil domains and a C-terminal region crucial for binding to other KMN network elements. It contains a PXVXL motif that allows interaction with HP1, which is critical for recruiting centromeric factors needed for proper chromosome segregation and mitotic fidelity. Although NSL1 is essential for cell division, it is not known as a direct therapeutic target, and no drugs currently interact with it or are designed to modulate its activity
