Overview

NTP-464 is a novel surface checkpoint protein and "multicellular orchestrator of inflammation resolution" identified by Nucleome Therapeutics through its proprietary 3D genome mapping platform. It is expressed on the surface of B cells and regulatory T cells (Tregs), where it plays a critical role in maintaining immune homeostasis. In patients with autoimmune diseases such as rheumatoid arthritis (RA), genetic variants in the non-coding "dark genome" lead to decreased expression of NTP-464 on B cells, contributing to dysregulated inflammation. Agonizing this target with a monoclonal antibody (also designated NTP-464) has been shown to inhibit B cell activation and proliferation while simultaneously enhancing the suppressive function of Tregs. This dual mechanism, which involves the induction of the anti-inflammatory enzyme IDO1, offers a first-in-class therapeutic approach for treating chronic inflammatory conditions including RA, systemic lupus erythematosus (SLE), and inflammatory bowel disease (IBD).
