Overview

Nuclear cap-binding protein subunit 1 (NCBP1, also known as CBP80) is a large protein component of the nuclear cap-binding complex (CBC) in eukaryotic cells[1][3][4]. The CBC binds to the 5' cap of newly synthesized pre-mRNA, playing a crucial role in mRNA biogenesis, including splicing, 3'-end formation, nuclear export, and protection from nuclease degradation[1][4][5]. NCBP1 itself does not bind RNA directly but stabilizes the CBC’s high-affinity binding through interaction with NCBP2 (CBP20) and recruits processing factors[1][3][4][5]. It is also an adaptor for alternative CBC formation with NCBP3 during stress conditions[5][7]. CBC-dependent processes managed by NCBP1 are essential for normal cell growth and viability; depletion leads to loss of cell proliferation and widespread misregulation of genes[5][7]. NCBP1 is not currently a therapeutic target or biomarker, and its dysfunction would pose major safety concerns due to pan-regulation of gene expression.
