Overview

Nuclear Deoxyribonucleic acid (nDNA) is the primary genetic material located within the cell nucleus, organized into complex structures called chromosomes (Wikipedia, 2024). It encodes the instructions necessary for the development, functioning, and reproduction of all living organisms. In the context of pharmacology, nuclear DNA is a classic therapeutic target, particularly in oncology, where cytotoxic drugs aim to disrupt its integrity or replication to halt the proliferation of malignant cells (NIH, 2023). Mechanisms include DNA alkylation, intercalation, and the inhibition of repair enzymes, which collectively lead to programmed cell death (PubMed, 2022). While highly effective against various cancers, targeting nuclear DNA often results in significant off-target effects because these drugs do not distinguish between the DNA of cancerous and healthy rapidly dividing cells (StatPearls, 2023). Modern approaches are shifting toward more specific genomic interventions, such as gene therapy and sequence-specific targeting, to improve the therapeutic index (Nature, 2021).
