POM121B possesses an N-terminal membrane-spanning domain, nuclear localization signals that facilitate nuclear targeting through importin-α/β, and C-terminal FG repeats that assist in molecular trafficking across the NPC[4][5]. It is indispensable for NPC assembly during interphase and interacts with scaffold NPC proteins such as Nup155 and Nup160[2][3]. POM121B has been implicated in cancer, both as a gene fusion partner and by mediating oncogenic transport into the nucleus[1].
