Overview

POM121C (Nuclear envelope pore membrane protein POM 121C) is a vertebrate-specific integral membrane nucleoporin, part of the nuclear pore complex (NPC) that anchors scaffold subcomplexes to the nuclear envelope via direct interaction with other nucleoporins (notably Nup155 and Nup160)[1]. Its N-terminal transmembrane domain and additional regions enable binding to the inner nuclear membrane and associated proteins (e.g., lamin B receptor)[2][3]. POM121 (and putatively POM121C) is indispensable for an early step in NPC assembly during interphase, marking assembly loci through interaction with the INM and serving as a scaffold for subsequent nucleoporin recruitment. Depletion of Pom121 results in reduced NPC number, nuclear pore clustering, and abnormal nuclear envelope structures, indicating its vital role in nuclear-cytoplasmic transport, nuclear architecture, and cell cycle progression. However, POM121C is not currently recognized as a direct drug target, has no known disease associations or biomarker utility, and is best considered a specialized structural component of the nuclear pore complex[1][2][3].
