Overview

Nuclear envelope pore membrane protein POM121 is an essential, single-pass type I transmembrane protein that serves as a core structural and functional component of the nuclear pore complex (NPC), a massive macromolecular complex embedded in the nuclear envelope[4][5][6]. POM121 anchors the NPC to the nuclear envelope through direct interactions with other nucleoporins, notably Nup155 and Nup160, facilitating both the assembly and structural integrity of the NPC[2]. Its N-terminal region spans the nuclear envelope membrane, while its C-terminal domain projects into the nucleoplasm and contains multiple FG-repeat motifs critical for nucleocytoplasmic transport[3]. Besides anchoring the NPC, POM121 contributes to the regulated transport of proteins and RNA between the nucleus and cytoplasm, and possesses additional roles in transcriptional regulation via soluble isoforms lacking the membrane anchor[1][3]. POM121 is involved in diverse cellular processes including cell cycle progression, gene regulation, and nuclear envelope reassembly after mitosis[1][3]. Disease associations include roles in cancer progression, where it can mediate nuclear import of oncogenic molecules, and in neurodegenerative diseases, inflammation, and viral infections such as HIV[1][3]. There are currently no approved drugs that directly target POM121, and it is not recognized as a therapeutic target in standard classification systems.
