Overview

XPO1 is a key nuclear export receptor responsible for transporting proteins and RNAs from the nucleus to the cytoplasm. It recognizes cargo proteins containing a leucine-rich nuclear export signal (NES) and forms a complex with these cargos and RanGTP in the nucleus. This complex translocates through the nuclear pore complex; once in the cytoplasm, GTP hydrolysis triggers release of cargo. XPO1 plays an essential role in maintaining cellular homeostasis. It is frequently overexpressed in multiple cancers and is associated with poor prognosis. Selective inhibitors of nuclear export (SINE) compounds have been developed to target XPO1.
