Overview

Nuclear factor I-B (NFIB) is a site-specific DNA-binding transcription factor belonging to the NFI family, crucial for the normal development of the lungs and the central nervous system. It regulates the expression of genes involved in cell differentiation and maturation, particularly during pulmonary alveolarization and glial cell fate specification. In clinical oncology, NFIB acts as a context-dependent regulator; it is a known driver of metastasis and tumor progression in small cell lung cancer (SCLC) and is frequently involved in the MYB-NFIB gene fusion characteristic of adenoid cystic carcinoma. Conversely, NFIB can function as a tumor suppressor in other malignancies, such as glioblastoma and bladder cancer, where its downregulation is associated with poor prognosis. Although direct pharmacological inhibition of NFIB remains a challenge due to its nature as a transcription factor, it is a significant target for therapeutic intervention and a promising biomarker for metastatic disease and patient stratification.
