Overview

Nuclear factor kappa B inhibitor zeta (NFKBIZ) mRNA encodes a nuclear IκB protein that serves as a critical regulator of the inflammatory response [UniProt Q9BYH8]. Unlike traditional IκB proteins that inhibit NF-κB by sequestering it in the cytoplasm, the protein translated from NFKBIZ mRNA is induced within the nucleus upon stimulation of Toll-like receptors or Interleukin-1 receptors [Yamamoto et al., 2004]. Once expressed, it binds to NF-κB p50 homodimers to selectively activate the transcription of pro-inflammatory cytokines such as IL-6 and IL-12p40 [Johansen et al., 2015]. NFKBIZ mRNA is significantly upregulated in autoimmune and inflammatory conditions, most notably psoriasis, where it mediates the pathogenic effects of the IL-17 pathway in keratinocytes [Mueller et al., 2018]. Therapeutic strategies targeting this mRNA, such as antisense oligonucleotides or siRNA, aim to specifically inhibit the production of downstream inflammatory mediators [NCBI Gene 64332]. Additionally, NFKBIZ dysregulation is a hallmark of certain B-cell lymphomas, suggesting that targeting its mRNA could have applications in oncology [Totzke et al., 2017].
