Overview

Nuclear Factor Kappa B Subunit 1 (NFκB1) is a key member of the NF-κB family of transcription factors. It is encoded by the NFKB1 gene and produces both a 105 kDa precursor protein (p105) and a 50 kDa processed form (p50). NFκB complexes regulate the transcription of over 200 genes involved in immune response, inflammation, cell survival, apoptosis, development, and stress responses. The most common active complex is the p65-p50 heterodimer. Activation occurs through the canonical or non-canonical pathways, ultimately leading to the translocation of NFκB dimers to the nucleus and binding to DNA. Dysregulation is associated with a wide range of diseases, including cancer, autoimmune disorders, and inflammatory conditions.

Mechanism of action

NFκB inhibitors prevent the translocation of NFκB dimers to the nucleus and/or their binding to DNA, thus inhibiting the expression of target genes.
