Overview

Nuclear factor kappa B subunit 1 (NFKB1) mRNA is the transcript encoding the p105 precursor protein, which is proteolytically processed into the p50 subunit of the NF-kappa-B transcription factor complex. NF-kappa-B is a master regulator of the immune response, inflammation, and cell survival, and its dysregulation is a hallmark of many chronic diseases. Overexpression of NFKB1 mRNA is frequently observed in various cancers and inflammatory conditions, where it drives the production of pro-inflammatory cytokines and promotes tumor cell survival. Consequently, NFKB1 mRNA has emerged as a therapeutic target for RNA-based modalities, such as small interfering RNAs (siRNAs) and antisense oligonucleotides (ASOs), which aim to reduce p50/p105 levels and dampen pathological signaling. However, therapeutic intervention must be carefully calibrated, as loss-of-function mutations in the NFKB1 gene are the most common monogenic cause of common variable immunodeficiency (CVID) in humans. Current research focuses on developing targeted delivery systems to minimize systemic toxicity and off-target effects while effectively modulating NF-kappa-B activity in specific tissues.
