Overview

Nuclear factor kappa B subunit 1 (p50) is a critical DNA-binding transcription factor belonging to the NF-κB family, encoded by the NFKB1 gene in humans[4][7][8]. It is generated by proteolytic processing of a larger precursor, p105, yielding the active p50 subunit which forms homo- or heterodimers, commonly pairing with RelA (p65) to create the classical NF-κB complex[1][3][5][6]. The p50-containing complexes regulate transcription of genes involved in immune and inflammatory responses, cell survival, and apoptosis, and mediate nuclear responses to a wide variety of cellular and environmental stimuli, including cytokines, stress, and infection[1][5][6][8]. Dysregulation of p50 and related NF-κB activity is implicated in several diseases, particularly cancer, chronic inflammatory disorders, and autoimmunity[5][6]. As a major mediator of immune signaling and inflammation, it is considered a validated and high-interest therapeutic target[1][5][8].
