Overview

Nuclear factor NF-kappa-B p100 subunit, encoded by the NFKB2 gene, is a key regulator of immune responses, inflammation, and cell survival. It exists as a precursor protein (p100) that is processed into the active transcription factor p52. NF-κB2 can function as both a transcriptional activator and repressor. Mutations in NFKB2 are associated with immunodeficiency syndromes like CVID and DAVID syndrome, and chromosomal rearrangements involving this locus are linked to B-cell lymphomas.

Mechanism of action

NF-κB2/p100 protein can act both as a transcriptional activator or repressor depending on its dimerization partner, regulating target gene expression. Also functions as an inhibitor by sequestering other NF-kappa-B subunits in the cytoplasm.
