Nuclear factor of activated T-cells, cytoplasmic 4 (NFATC4) is a calcium/calmodulin-activated transcription factor in the NFAT family, regulated by the phosphatase calcineurin. It is primarily involved in immune function, controlling inducible expression of multiple cytokine genes (notably IL-2 and IL-4) following T cell activation. Beyond immune cells, NFATC4 plays important roles in cardiac development, neuronal survival, cell differentiation (including adipogenesis), and mitochondrial function. Its activity is regulated by nuclear translocation upon dephosphorylation and can be inhibited by immunosuppressant drugs such as cyclosporin A and tacrolimus, conferring clinical relevance both as a drug target and as a contributor to adverse effects of immunosuppressive therapy[1][2][3][5].
