Nuclear factor of kappa light polypeptide gene enhancer in B-cells inhibitor delta (NFKBID)

Target
NFKBID
Molecular classification
Transcription regulator (NF-κB inhibitor), Nuclear IκB protein
01

Overview

Nuclear factor of kappa light polypeptide gene enhancer in B-cells inhibitor delta (NFKBID) is a nuclear protein of 327 amino acids belonging to the atypical nuclear inhibitors of NF-κB. It regulates NF-κB transcription factor activity, impacting immune responses, germinal center formation, and the development and function of B cells, plasma cells, and certain T cell populations. NFKBID functions as both inducer and inhibitor of NF-κB-dependent gene expression and is required for immunosuppressive regulatory T cell generation. Its deficiency or dysfunction has been linked to immune dysregulation and deficiency syndromes.

Mechanism of action

Drugs affecting downstream NF-κB signaling may modulate NFKBID activity indirectly (activation or inhibition of NF-κB cascade)

03

Biological functions

Regulation of NF-κB signalingPromotion of germinal center reactionsImmune system homeostasisRegulation of B cell and plasma cell function/developmentRegulation of T helper 17 cell differentiationRegulation of cytokine expression (e.g., IL-2, IL-6)Inflammatory response
04

Disease associations

Immune deficiency (e.g., ectodermal dysplasia and immune deficiency)InflammationImmunological disordersPotential involvement in infection (via NF-κB pathway)
05

Safety considerations

Potential risks involve immunosuppression or unwanted immune activation due to disruption of NF-κB regulation; no specific issues reported for NFKBID
06

Interacting drugs

None directly reported for NFKBID; many known drugs target the NF-κB pathway but not specifically NFKBID
07

Biomarkers

None reported specifically for NFKBID; NF-κB activity in general is monitored using cytokine levels, but not NFKBID protein directly
