Overview

Nuclear factor related to kappa-B-binding protein (NFRKB) is a protein-coding gene encoding a metazoan-specific subunit of the INO80 chromatin remodeling complex. NFRKB binds specific DNA sequences (5'-GGGGAATCTCC-3'), and acts as a regulatory component that modulates transcriptional regulation, DNA replication, and repair—especially telomere maintenance and stability. In cancer biology, particularly hepatocellular carcinoma, NFRKB is highly expressed and accumulates in tumor nuclei but is lower in normal hepatocytes. Its increased levels correlate with poor clinical outcomes. Pathway analysis links NFRKB to transcription-coupled nucleotide excision repair and protein metabolism. NFRKB also influences the activity of the deubiquitinase UCHL5 within the INO80. No drugs currently modulate NFRKB directly, but its prognostic value highlights its biological and therapeutic interest, particularly in oncology
