Overview

Nuclear fragile X mental retardation-interacting protein 1 (NUFIP1) is a conserved RNA-binding protein that shuttles between the nucleus and cytoplasm. It interacts with fragile X mental retardation protein (FMRP), is present in the nuclear matrix, and is also found with ribosomes in the cytoplasm, particularly in neurons. NUFIP1 is involved in the export and localization of mRNA and regulates local protein synthesis at synapses. Notably, it acts as an autophagy receptor for ribosome degradation during starvation-induced ribophagy by binding directly to ribosomes and LC3B on autophagosomes. NUFIP1 also participates in the assembly of box C/D small nucleolar ribonucleoprotein particles (snoRNPs), and its loss affects ribosome homeostasis and the response to nutrient deprivation. There are currently no drugs targeting NUFIP1, and it is not considered a conventional therapeutic target such as a receptor or enzyme.
