Overview

Nuclear receptor 2C2-associated protein (NR2C2AP), also known as TRA16, is a nuclear protein encoded by the NR2C2AP gene. It acts as a corepressor of Testicular orphan nuclear receptor 4 (TR4, also called NR2C2) by binding to TR4 and selectively inhibiting its ability to bind DNA response elements and activate target gene transcription. NR2C2AP/TRA16 is predominantly localized in the nucleus, contains 139 amino acids (16 kDa), and shows specificity as a corepressor for TR4, exerting minimal effects on androgen, glucocorticoid, or progesterone receptor transactivation. Its function appears to be the selective repression of TR4-mediated activities, which potentially impact cell signaling in specific tissues, such as testis and possibly in cancer cell contexts. No drugs are directly known to target NR2C2AP, and it is not classified as a canonical therapeutic target.
