Nuclear receptor-binding factor 2 (NRBF2) is a multifunctional protein that acts as a tightly bound fifth subunit of the class III phosphatidylinositol 3-kinase complex I (PI3KC3-C1), central to autophagy initiation and regulation. NRBF2 directly enhances VPS34 lipid kinase activity and stabilizes PI3KC3-C1 assembly by interacting with subunits ATG14 and Beclin 1. It is also known to modulate transcription by interacting with nuclear receptors, and participates in synaptic plasticity and learning/memory processes in neurons, possibly through autophagy-independent mechanisms as well. NRBF2 has roles in neurodevelopment, autophagosome maturation, and is implicated in cognitive disorders, Alzheimer’s disease, and other neurodegenerative conditions. Loss or dysfunction of NRBF2 impairs autophagy and memory acquisition, and has been linked to disease phenotypes including dystonia and optic atrophy.
