Overview

Nuclear receptor-binding protein 1 (NRBP1) is a pseudokinase and adaptor protein that plays diverse roles in cellular biology[3][1]. It is implicated in embryonic development, subcellular trafficking (especially ER-to-Golgi transport), and regulation of cell fate and proliferation, notably in the intestinal epithelium[1]. In cancer biology, its function is context-dependent: NRBP1 acts as a scaffold for the P-Rex1/Rac1/Cdc42 signaling axis, promoting migration, invasion, and proliferation in triple-negative breast cancer cells, and may serve as a context-specific tumor suppressor or oncogene[2][3]. NRBP1 also participates in proteasome-mediated protein degradation, acting as a substrate recognition factor in certain E3 ubiquitin ligase complexes[4][3]. Interactions with viral proteins (e.g., Dengue NS3) suggest roles in host-pathogen interactions, potentially altering intracellular membrane architecture to facilitate viral replication[1]. Although important for several cellular functions and cancer pathways, NRBP1 is not currently an established therapeutic drug target.
